    Augmented Reality to Engage Visitors of Science Museums through Interactive Experiences

    In the last years, interactive exhibitions based on digital technologies have become widely common, thanks to their flexibility and effectiveness in engaging visitors and creating memorable experiences. One of the topics in which digital technologies can be particularly effective is the communication of abstract concepts that are difficult for the human mind to imagine. An emblematic example is the astronomy discipline, which requires us to imagine and understand phenomena far away from our everyday life. In this paper, the authors present a research project, MARSS, in which digital technologies are used effectively to enhance the Users' Experience of the Museo Astronomico di Brera located in Milan. Specifically, the MARSS project aims at designing and developing a new digital journey inside the museum to allow different categories of visitors to enjoy the exhibition in an engaging and interactive way. The paper presents the design and development phases of the experience and its evaluation with users. The results of the evaluation indicate that the digital interactive experience is appreciated by users and is successful in translating the content of high scientific value into more engaging and easily understandable elements

    Geoelectrical characterization and monitoring of slopes on a rainfall-triggered landslide simulator

    In this paper, we present the results of time-lapse electrical resistivity tomography (ERT) monitoring of rainfall-triggered shallow landslides reproduced on a laboratory-scale physical model. The main objective of our experiments was to monitor rainwater infiltration through landslide body in order to improve our understanding of the precursors of failure. Time-domain reflectometry (TDR) data were also acquired to obtain the volumetric water content. Knowing the porosity, water saturation was calculated from the volumetric water content and we could calibrate Archie's equation to calculate water saturation maps from inverted resistivity values. Time-lapse ERT images proved to be effective in monitoring the hydrogeological conditions of the slope as well as in detecting the development of fracture zones before collapse. We performed eight laboratory tests and the results show that the landslide body becomes unstable at zones where the water saturation exceeds 45%. It was also observed that instability could occur at the boundaries between areas with different water saturations. Our study shows that time-lapse ERT technique can be employed to monitor the hydrogeological conditions of landslide bodies and the monitoring strategy could be extended to field-scale applications in areas prone to the development of shallow landslides

    Improved Molecular Diagnosis of McCune-Albright Syndrome and Bone Fibrous Dysplasia by Digital PCR

    McCune-Albright syndrome (MAS) is a rare congenital disorder characterized by the association of endocrine and nonendocrine anomalies caused by somatic activating variants of GNAS. The mosaic state of variants makes the clinical presentation extremely heterogeneous depending on involved tissues. Biological samples bearing a low level of mosaicism frequently lead to false-negative results with an underestimation of causative molecular alterations, and the analysis of biopsies is often needed to obtain a molecular diagnosis. To date, no reliable analytical method for the noninvasive testing of blood is available. This study was aimed at validating a novel and highly sensitive technique, the digital PCR (dPCR), to increase the detection rate of GNAS alterations in patients with a clinical suspicion of MAS and, in particular, in blood. We screened different tissues (blood, bone, cutis, ovary, and ovarian cyst) collected from 54 MAS patients by different technical approaches. Considering blood, Sanger was unable to detect mutations, the allele-specific PCR and the co-amplification at lower denaturation temperature had a 9.1% and 18.1% detection rate, respectively, whereas the dPCR reached a 37.8% detection rate. In conclusion, the dPCR resulted in a cost-effective, reliable, and rapid method allowing the selective amplification of low-frequency variants and able to improve GNAS mutant allele detection, especially in the blood

    Minding the gap between cortisol levels measured with second-generation assays and current diagnostic thresholds for the diagnosis of adrenal insufficiency: a single-center experience

    Purpose: The current cut-offs for the diagnosis of adrenal insufficiency (AI) have been established using outdated immunoassays. We compared the cortisol concentrations measured with Roche Cortisol I (R1), the newly available Roche Cortisol II (R2), and liquid chromatography tandem mass spectrometry (LC-MS/MS), the gold standard procedure to measure steroids in patients undergoing the corticotropin (ACTH) test. Methods: We enrolled 30 patients (age 47 \ub1 21 years) referred to undergo the ACTH test (1 or 250 \u3bcg). Cortisol was measured at 0, 30, and 60 min after stimulation with R1, R2, and LC-MS/MS. AI was diagnosed for R1-stimulated peak cortisol concentrations < 500 nmol/L. Results: Mean cortisol concentrations measured with R2 and LC-MS/MS were comparable, while mean cortisol concentrations measured by R1 were higher than those of both R2 and LC-MS/MS (respectively, basal 411 \ub1 177, 287 \ub1 119, and 295 \ub1 119 nmol/L; at 30 min, 704 \ub1 204, 480 \ub1 132, and 500 \ub1 132 nmol/L; at 60 min, 737 \ub1 301, 502 \ub1 196, and 519 \ub1 201 nmol/L, p 64 0.01 for R1 vs. both R2 and LC-MS/MS at each point). Considering the 500 nmol/L cortisol peak cut-off, AI was diagnosed in 5/30 patients using R1 and in 12/30 using R2 (+ 140%). Based on the correlation between R1 and R2, the threshold of 500 nmol/L became 351 nmol/L (12.7 \u3bcg/dL) when cortisol was measured with R2, and 368 nmol/L (13.3 \u3bcg/dL) with LC-MS/MS. Conclusions: The use of more specific cortisol assays results in lower cortisol concentrations. This could lead to misdiagnosis and overtreatment when assessing AI with the ACTH test if a different cut-off for cortisol peak is not adopted

    Cyclic RGD peptidomimetics containing bifunctional diketopiperazine scaffolds as new potent integrin ligands

    The synthesis of eight bifunctional diketopiperazine (DKP) scaffolds is described; these were formally derived from 2,3-diaminopropionic acid and aspartic acid (DKP-1-DKP-7) or glutamic acid (DKP-8) and feature an amine and a carboxylic acid functional group. The scaffolds differ in the configuration at the two stereocenters and the substitution at the diketopiperazinic nitrogen atoms. The bifunctional diketopiperazines were introduced into eight cyclic peptidomimetics containing the Arg-Gly-Asp (RGD) sequence. The resulting RGD peptidomimetics were screened for their ability to inhibit biotinylated vitronectin binding to the purified integrins \u3b1 v\u3b2 3 and \u3b1 v\u3b2 5, which are involved in tumor angiogenesis. Nanomolar IC 50 values were obtained for the RGD peptidomimetics derived from trans DKP scaffolds (DKP-2-DKP-8). Conformational studies of the cyclic RGD peptidomimetics by 1H NMR spectroscopy experiments (VT-NMR and NOESY spectroscopy) in aqueous solution and Monte Carlo/Stochastic Dynamics (MC/SD) simulations revealed that the highest affinity ligands display well-defined preferred conformations featuring intramolecular hydrogen-bonded turn motifs and an extended arrangement of the RGD sequence [C\u3b2(Arg)-C\u3b2(Asp) average distance 658.8 \uc5]. Docking studies were performed, starting from the representative conformations obtained from the MC/SD simulations and taking as a reference model the crystal structure of the extracellular segment of integrin \u3b1 v\u3b2 3 complexed with the cyclic pentapeptide, Cilengitide.     The census of nuclear activity of late-type galaxies in the Virgo cluster

    The first spectroscopic census of AGNs associated to late-type galaxies in the Virgo cluster is carried on by observing 213 out of a complete set of 237 galaxies more massive than M_dyn>10^{8.5} solar masses. Among them, 77 are classified as AGNs (including 21 transition objects, 47 LINERs and 9 Seyferts), and comprize 32% of the late-type galaxies in Virgo. Due to spectroscopic incompleteness at most 21 AGNs are missed in the survey, so that the fraction would increase up to 41%. Using corollary Near-IR observations, that enable us to estimate galaxies dynamical masses, it is found that AGNs are hosted exclusively in massive galaxies, i.e. M_dyn\gsim 10^{10} solar masses. Their frequency increases steeply with the dynamical mass from zero at M_dyn\approx10^{9.5} solar masses to virtually 1 at M_dyn>10^{11.5} solar masses. These frequencies are consistent with the ones of low luminosity AGNs found in the general field by the SDSS. Massive galaxies that harbor AGNs commonly show conspicuous r-band star-like nuclear enhancements. Conversely they often, but not necessarily contain massive bulges. Few well known AGNs (e.g. M61, M100, NGC4535) are found in massive Sc galaxies with little or no bulge. The AGN fraction seems to be only marginally sensitive to galaxy environment. We infer the black hole masses using the known scaling relations of quiescent black holes. No black holes lighter than ∼106\sim 10^6 \msol are found active in our sample.Comment: The paper contains 13 figures and 5 tables; accepted for publication in MNRA
